Combine sugar, butter, and milk in a 3 qrt glass bowl. Cook in microwave, uncovered, 5 1/2 to 6 1/2 minutes, or until mixture comes to a boil, stirring once or twice. Cook uncovered for 3 more minutes, stirring occasionally if necessary to keep mixture from boiling over. Stir in peanut butter, marshmallow cream, and vanilla until well-blended. Pour into battered pan. Refrigerate 2 hours; cut into squares.
