In a large mixing bowl, cream the butter and sugar with a mixer until light and fluffy. Beat in the eggs, one at a time, until creamy. Beat in milk, lemon zest, vanilla and rum.
In a separate bowl, add all but 1 TBS of the flour. Add the baking powder and salt, stir to combine. Gradually beat into the egg mixture a little bit at a time, beating until each addition is fully incorporated.
Toss the blueberries with the remaining 1 TBS of flour. Gently fold half of the berries into the cake batter.
Prepare a loaf pan by spraying with non-stick spray. Cover the bottom with parchment paper. Add half of the cake batter to the pan. Sprinkle with the reserved blueberries, and then top with the remaining cake batter, smoothing the top.
Bake at 350’ for about 1 hour and 20-30 minutes, or until a toothpick inserted into the center comes out clean. Remove from the oven and prepare the glaze.
Combine the sugar and lemon juice in a microwave-safe container. Microwave on high for 30 seconds, stirring once, until the sugar is dissolved.
Using a bamboo skewer, poke holes all over the top of the cake. Drizzle the syrup over the cake and let cool in the pan on a wire rack for 10 minutes.
Remove from pan and let cool completely. Store tightly covered with plastic wrap at room temperature.
